Using nano “couriers” to deliver PKD drugs to just the right address

The term “mRNA lipid nanoparticle” has become common of late, given the use of Pfizer/BioNTech’s and Moderna’s COVID-19 vaccines make of nanoparticles to carry mRNA into cells. Once inside a cell, the mRNA instructs the cell to make pieces of the proteins that make up the SARS-CoV-2 virus, a process called “translation.” In so doing, […] Find jobs at Careers by KevinMD.com. Search thousands of physician, PA, NP, and CRNA jobs now. Learn more.

CS Soapbox: The Novel That Inspired Matrix, Inception & WandaVision

BEGIN SLIDESHOW CS Soapbox: The Novel That Inspired Matrix, Inception & WandaVision In 1969 the famed sci-fi author Philip K. Dick wrote what was arguably his most groundbreaking and influential novel, titled Ubik. Since his death in 1982 the late writer’s work has become a cornerstone of the genre (the phrase “Dickian” is common parlance) and has been transformed into numerous classic films and television (Blade Runner, Minority Report, Total Recall, The Man in the High Castle), yet Ubik...

Study describes the diversity of genetic changes that cause inherited kidney disease

A study has described genetic changes in patients with the most common form of hereditary kidney disease that affects an estimated 12.5 million people worldwide. The research, which focussed on Polycystic Kidney Disease (PKD) in Ireland, provides insights into PKD that will assist doctors and patients in the management of this of inherited condition.

Genetic Discrimination Measures Upheld

In Chapter 11 of the “Canadian Health Law Practice Manual,” Genetics and the Law, Amy Zarzeczny, Tracey M. Bailey and Timothy Caulfield state, One concern that consistently emerges in relation to obtaining genetic information is the worry that an individual may be discriminated against on the basis of his or her genetic make-up and, specifically, on the basis of a predisposition to a certain condition or disease. Discrimination can of course occur on a wide variety of fronts including, but not l...

Crystal deposition triggers tubule dilation that accelerates cystogenesis in polycystic kidney disease

The rate of disease progression in autosomal-dominant polycystic kidney disease (ADPKD) has high intrafamilial variability, suggesting that environmental factors may play a role. We hypothesized that a prevalent form of renal insult may accelerate cystic progression and investigated tubular crystal deposition. We report that calcium oxalate (CaOx) crystal deposition led to rapid tubule dilation, activation of PKD-associated signaling pathways, and hypertrophy in tubule segments along the affecte...

Structure of the human PKD1-PKD2 complex

Mutations in two genes, PKD1 and PKD2, account for most cases of autosomal dominant polycystic kidney disease, one of the most common monogenetic disorders. Here we report the 3.6-angstrom cryo–electron microscopy structure of truncated human PKD1-PKD2 complex assembled in a 1:3 ratio. PKD1 contains a voltage-gated ion channel (VGIC) fold that interacts with PKD2 to form the domain-swapped, yet noncanonical, transient receptor potential (TRP) channel architecture. The S6 helix in PKD1 is broken ...

2018 Porsche 911 GT3 First Test: Beauty Beheld

I stopped counting at 11, any significance to the number a happy coincidence. I always record my conversations with Randy Pobst, held in the doorway of whatever car he’s just lapped. It’s best to get him while everything’s fresh, before anyone says anything that might color his impressions. Today, his impressions were things of beauty. “There’s very little to complain about,” he said. “It behaves so beautifully. The balance is so beautiful. You still have to be a little bit careful not going ...

Neither Huxleyed, nor Orwelled: living in the Phildickian dystopia

Political scientist and sf fan Henry Farrell (previously) argues persuasively that the dystopian elements of our everyday life are best viewed through the lens of Philip K Dick (whose books repeatedly depicted a world of constructed realities, whose true nature was obscured by totalitarians, conspiracies, and broken computers) and not Orwell or Huxley, whose computers and systems worked altogether too well to be good parallels for today's janky dystopia. (more…)

Integrated field, laboratory, and theoretical study of PKD spread in a Swiss prealpine river [Ecology]

Proliferative kidney disease (PKD) is a major threat to wild and farmed salmonid populations because of its lethal effect at high water temperatures. Its causative agent, the myxozoan Tetracapsuloides bryosalmonae, has a complex lifecycle exploiting freshwater bryozoans as primary hosts and salmonids as secondary hosts. We carried out an integrated...

Lysine methyltransferase SMYD2 promotes cyst growth in autosomal dominant polycystic kidney disease

Autosomal dominant polycystic kidney disease (ADPKD) is driven by mutations in PKD1 and PKD2 genes. Recent work suggests that epigenetic modulation of gene expression and protein function may play a role in ADPKD pathogenesis. In this study, we identified SMYD2, a SET and MYND domain protein with lysine methyltransferase activity, as a regulator of renal cyst growth. SMYD2 was upregulated in renal epithelial cells and tissues from Pkd1-knockout mice as well as in ADPKD patients. SMYD2 deficiency...

If Rigs Counts Are Rising, Why Did These Rig Stocks Decline in March?

Despite the improving environment for rigs in the U.S., land drillers took a nosedive in March. Shares of Patterson-UTI Energy (NASDAQ: PTEN) , Nabors Industries (NYSE: NBR) , Parker Drilling (NYSE: PKD) , and Precision Drilling (NYSE: PDS) all declined double digits last month. Wasn't the onshore rig industry supposed to be in the middle of a revival? According to Baker Hughes' weekly rig count survey, active rigs in the field improved 10% in March to a total count of 824. Throughout 201...

Oil and Gas Stock Roundup: Look Out Below!

Philip K. Dick’s Electric Dreams on Amazon

Early this morning I saw an email from Dave that pointed me at a giant bundle of happiness. Bryan Cranston Will Bring Philip K. Dick’s Electric Dreams to Series So – let’s go through all the parts of awesomeness of this. First, Philip K. Dick is one of my favorite sci-fi writers. One summer, about five or so years ago, I bought every PKD book I could find in paperback (almost all of them.) I started reading them from first to last. I’ve gotten through about half of them and have sporadically rea...

Book: The Man in the High Castle

I had a great digital sabbath yesterday with Amy and my friends Dave and Maureen. I had a cold so I took a three hour nap in the middle of the day. For the rest of the day, I sat around in my PJs. I read Philip K. Dick’s The Man in the High Castle. Amy and I loved Season 1 of the Amazon series but struggled with the first episode on Season 2 (I’m sure we’ll fight through it and watch it this week.) Since we got off to a slow start on Season 2, I decided to read the book, which I had resisted rea...

APEX Wins the Philip K. Dick Award

Tonight, in Seattle, I was in the crowd when my novel Apex won the Philip. K Dick Award. Apex is the third and final book of the Nexus Trilogy. Those books have now collectively won the Prometheus Award, the Endeavor Award, been listed on NPR’s list of Best Books of the Year, and been shortlisted for the Arthur C. Clarke Award and the Kitchies Golden Tentacle Award. They also earned me a nomination for the Campbell Award for Best New Author in 2014.To say I’m pleased would be an understatement....
